Our work aims to purify, characterize and evaluate a laccase from by-products of the shrimp farming industry (Litopenaeus vannamei) for the degradation of Polycyclic Aromatic Hydrocarbons (PAHs) from 2019 oil spill in Brazilian coast. The enzyme was purified by affinity chromatography and characterized as thermostable, with activity above 90 °C and at alkaline pH. In addition, the laccase was also tolerant to copper, lead, cadmium, zinc, arsenic, hexane and methanol, with significant enzymatic activation in acetone and 10 mM mercury. Concerning PAHs' degradation, the enzyme degraded 42.40 % of the total compounds, degrading >50 % of fluorene, C4-naphthalenes, C3-naphthalenes, C2-naphthalenes, anthracene, acenaphthene, 1-methylnaphthalene and 2-methylnaphthalene. Thus, this laccase demonstrated important characteristics for bioremediation of marine environments contaminated by crude oil spills, representing a viable and ecological alternative for these purposes.

INTRODUCTION: Coconut oil has been considered as a therapeutic alternative in several pathologies, but there is limited information regarding its effects on brain functioning. OBJECTIVE: This study analyzed whether early virgin coconut oil (VCO) supplementation interferes with electrical activity of the adult rat brain and its lipid peroxidation. Moreover, it investigated whether the putative effect on brain electrophysiology could be affected by overnutrition occurring during lactation, and/or by environmental enrichment (EE). Electrophysiology was measured through cortical spreading depression (CSD), a phenomenon related to brain excitability. METHODS: Wistar rats were suckled in litters of either nine or three pups, forming nourished (N) or overnourished (ON) groups, respectively. Between the 7th and 30th days of life, half of the animals in each group received VCO (10 mg kg-1 d-1; by gavage). The other half received an equivalent amount of vehicle (V, 0.009% cremophor). On day 36, animals from both groups were subjected to EE for 4 weeks. At 105 ± 15 days of life, each animal was subjected to CSD recordings and lipid peroxidation analyses. RESULTS: Overnutrition during lactation enhanced body and brain weights. VCO decelerated the CSD propagation velocity (control - 3.57 ± 0.23 mm min-1versus VCO - 3.27 ± 0.18 mm min-1; p < 0.001), regardless of whether subjected to overnourishment or EE exposure. Neither VCO nor EE modified the cerebral lipid peroxidation (p > 0.05). CONCLUSION: VCO supplementation impaired the spreading of CSD, indicating reduction of brain excitability. VCO effects occurred regardless of the nutritional state during lactation.

Sepsis is a clinical syndrome with high morbidity and mortality. It is characterized by acute inflammatory response and oxidative stress, which is implicated in cerebral dysfunction. Murici (Byrsonimacrassifolia (L.) Kunth) is a fruit rich in antioxidant compounds, which could be an alternative to prevent damage to tissues induced by sepsis . Here, we evaluated the effects of sepsis on the propagation of cortical spreading depression (CSD) and oxidative stress, and tested the action of murici antioxidant extract in prevention against the effect of sepsis. Male Wistar rats (90-210 days, n = 40) were previously supplemented, orogastrically, with murici extract (150 mg/kg/day or 300 mg/kg/day), or an equivalent volume of the vehicle solution, for fifteen days. Then the animals were subjected to experimental sepsis through cecal ligation and perforation (CLP). Subsequently, CSD recordings were obtained and brain oxidative stress was evaluated. Sepsis decelerated CSD and increased the malondialdehyde (MDA) levels in the brain cortex of the animals. In contrast, septic rats that had been previously supplemented with murici antioxidant extract in doses of 150 and 300 mg/kg/day showed an increase in CSD propagation velocity, low levels of MDA and GSH/GSSG ratio and an increase of superoxide dismutase (SOD) activity, regardless of the dose tested. Our results demonstrate that sepsis affects brain excitability and that this effect can be prevented by murici antioxidant extract. The effects of sepsis and/or murici extract on CSD may be due to the oxidative state of the brain.

Objectives: This investigation studied whether physical exercise could modulate cortical spreading depression (CSD) propagation velocity in adult rat offspring from dams that had received a high-fat (HF) diet during lactation. Methods: Wistar male rats suckled by dams fed either control (C) or HF diet ad libitum. After weaning, pups received standard laboratory chow. From 40 to 60 days of life, half of the animals exercised on a treadmill (group E); the other half remained sedentary (group S). Two additional HF groups (E and S) received fluoxetine (F; 10 mg/kg/day, orogastrically) from 40 to 60 days of life (groups HF/EF and HF/F). Results: At 40 days of life, rats from the maternal HF diet presented higher weight, thoracic circumference, and Lee Index than C animals and remained heavier at 60 days of life. Physical exercise decreased abdominal circumference. HF diet increased CSD propagation velocity (mean ± SD; mm/min) in sedentaries (HF/S 3.47 ± 0.31 versus C/S 3.24 ± 0.26). Treadmill exercise decelerated CSD propagation in both groups C/E (2.94 ± 0.28) and HF/E (2.97 ± 0.40). Fluoxetine alone decreased CSD propagation (HF/F 2.88 ± 0.45) compared with HF/S group. The combination of fluoxetine + exercise under HF condition (2.98 ± 0.27) was similar to HF/E group. Discussion: Physical exercise is able to reduce CSD propagation velocity in rat adult brains even when they have suffered over-nourishing during lactation. The effects of exercise alone or fluoxetine alone on CSD were similar to the effects of fluoxetine + exercise, under the HF condition. Data reinforce malnutrition during lactation modifies cortical electrophysiology even when the HF condition no longer exists.

The EE decelerated CSD propagation velocity regardless of nutritional state during lactation (p < 0.001). The CSD deceleration in the N-EE group was 23.8% and in the ON-EE group was 15% in comparison with the N-SE and ON-SE groups, respectively. CONCLUSION: Our data demonstrated that EE exposure in the juvenile phase of the rat's life reduced brain excitability, and this effect was observed even if animals were overnourished during lactation. An EE could be considered an adjuvant therapeutic resource to modulate brain excitability.

RESUMO Objetivo: Este estudo analisou se a exposição ao ambiente enriquecido durante a fase juvenil da vida interferiria na atividade elétrica do cérebro de ratos adultos. Além disso, a presente pesquisa também investigou se esse provável efeito na atividade elétrica cerebral poderia ser afetado pela hipernutrição durante a lactação. A eletrofisiologia foi medida através da depressão alastrante cortical, um fenômeno relacionado à excitabilidade cerebral. Métodos: Ratos Wistar foram amamentados em ninhadas de nove ou três filhotes, formando os grupos nutridos ou hipernutridos, respectivamente. Aos 36 dias, metade dos animais de cada condição nutricional foram expostos ao ambiente enriquecido. A outra metade foi mantida na condição de ambiente padrão. Aos 90-120 dias de vida, foram obtidos os registros da depressão alastrante cortical. Resultados: A hipernutrição durante a lactação causou incrementos (p < 0,05) nos pesos corporal e cerebral.O Ambiente Enriquecido desacelerou a velocidade de propagação da depressão alastrante cortical independentemente do estado nutricional durante a lactação (p < 0,001). A desaceleração da depressão alastrante cortical no grupo nutrido/ambiente enriquecido foi de 23,8% e no grupo hipernutrido/ambiente enriquecido foi de 15% em comparação com os grupos nutrido/ambiente padrão e hipernutrido/ambiente padrão, respectivamente. Conclusão: Nossos dados demonstram que a exposição ao ambiente enriquecido na fase juvenil da vida do rato reduz a excitabilidade cerebral, e esse efeito pode ser observado mesmo se os animais estiverem hipernutridos durante a lactação. O ambiente enriquecido pode ser considerado um recurso terapêutico adjuvante para modular a excitabilidade cerebral.

PURPOSE: To investigate the effects of Murici extract on the brain excitability-dependent phenomenon known as cortical spreading depression (CSD) and on brain oxidative stress. METHODS: Adult and aged Wistar rats were supplemented with murici extract (150 mg/kg/day or 300 mg/kg/day) by gavage for fifteen days. Afterwards, the animals were submitted to a CSD electrophysiological recording and to brain oxidative stress evaluation. RESULTS: Our results showed that aging decreased CSD propagation velocity, catalase activity and glutathione/oxidized glutathione ratio (GSH/GSSG) in the brain cortex of the rats, and increased malondialdehyde (MDA) concentrations and superoxide dismutase (SOD) activity. The highest dose (300 mg/kg/day) of murici extract accelerated CSD, whereas the lowest (150mg/kg/day) decelerated, in both adult and aged animals. In contrast, aged animals supplemented with murici extract in both doses presented low MDA levels and high GSG/GSSG ratio in comparison to the control-aged animals. CONCLUSION: Murici extract supplementation seems to revert detrimental effects in aged brains and could be considered as a strategy in the treatment of pathologies related to aging and cortical spreading depression.

PURPOSE: To investigate the effect of kefir and banana pulp and skin flours on the serum levels of total cholesterol, HDL-c, LDL-c and triacylglycerols in rats fed cholesterol-rich diet. METHODS: Thirty Male Wistar rats were used. In the first 21 days, the animals were fed purified hypercholesterolemic diets, except the standard group. In the next 21 days, the animals were given modified diets: Group GC: standard diet AIN-93G; Group HIP: hypercholesterolemic diet; Group F: hypercholesterolemic diet added of 1% of banana skin flour and 7% of banana pulp flour; Group Q: hypercholesterolemic diet plus kefir suspension by oral infusion (1.5 ml/animal); Group FQ: hypercholesterolemic diet added of 1% banana skin flour and 7% of banana pulp flour plus kefir suspension (1.5 ml/animal). RESULTS: In spite of the high fiber content, the addition of banana pulp (7%) and skin (1%) flour did not alter the plasma levels of total cholesterol, HDL-c and LDL-c. However, they reduced the TG levels in 22%. Already fermented kefir reduced significantly the levels of VLDL, LDL-c and triacylglycerols, in addition to having increased HDL-c. However, it was not possible to verify the symbiotic effect between both. CONCLUSION: The results reinforce the beneficial effects of kefir in reducing the risks of cardiovascular diseases.

Introdução: O murici (Byrsonima crassifolia (L.) Kunth) é um fruto utilizado pela população como alimento ou como agente terapêutico, mas ainda há poucos estudos sobre as suas propriedades funcionais. Assim, este trabalho objetivou caracterizar os frutos do murici quanto à composição de compostos antioxidantes e avaliar seus mecanismos de ação antioxidante in vitro. Metodologia: Os frutos do murici foram coletados na cidade de Araiozes, Maranhão, Brasil. A composição centesimal foi avaliada pelos métodos oficiais de análise e o perfil de minerais por espectrometria de emissão ótica com plasma indutivamente acoplado. Os perfis de carotenóides, tocoferóis e vitamina C foram determinados por comatografia líquida de alta eficiência. As condições ótimas para a extração de polifenóis de murici foram definidas por meio de planejamento composto central rotacional. Assim, o extrato A foi obtido usando 43,4 por cento de acetona a 28,9°C por 50,8 minutos e o extrato B com 45,2 por cento de acetona a 40°C por 52,8 minutos. Os extratos foram avaliados quanto ao perfil de compostos fenólicos por HPLC-ESI/MS e quanto aos mecanismos de ação antioxidante in vitro. Resultados: Os frutos de murici apresentaram (valor médio) 74,35 por cento de umidade, 0,94 por cento de cinzas, 0,68 por cento de proteínas, 1,82 por cento de lipídios, 4,31 por cento de carboidratos disponíveis e 17,91 por cento de fibras. Quanto ao perfil de minerais, o murici é uma boa fonte de potássio (338.58mg 100 g 1 ), cobre (200 µg 100g ) e magnésio (35.91 mg 100 g 1 ). Os frutos apresentaram 57,41 mg/100 g vitamina C, 308,97 µg/g de luteína, 16,78 µg/g de -caroteno, 3,80 µg/g de -criptoxantina, 6,49 mg/100 g de - tocoferol, 017 mg/100 g de -tocoferol, 2,66 mg/100 g de -tocoferol e 0,33 mg/100 g de -tocoferol. A análise por HPLC-ESI/MS permitiu a identificação de 19 compostos fenólicos nos extratos de murici, sendo três dímeros de proantocianidinas, dois isômeros de catequina, ácido gálico, quercetina e seus derivados, entre outros. Os extratos de murici agiram eficientemente contra os radicais ABTS (TEAC de 158,48 µM Trolox/ g murici fresco para o extrato A e 170,17 para o extrato B), peroxil (1252,88 µM Trolox/ g extrato para o extrato A e 1332,78 para o extrato B), hidroxil (IC = 300,68 µg/mL para o extrato A e 281,33 para o extrato B), superóxido (IC 50 = 374,50 µg/mL para o extrato A e 350,92 para o extrato B)

O Brasil é um dos países que mais produz resíduos agroindustriais, como os resíduos de frutas pelas indústrias de polpas, o que tem contribuído para o aumento da produção do lixo orgânico, provocando graves problemas ambientais. Nesse contexto, estudos têm sido conduzidos com o intuito de investigar o valor nutricional desses resíduos, valorizando-os e sugerindo novas alternativas de utilização. Assim, neste trabalho, objetivou-se realizar a caracterização nutricional e determinar os compostos antioxidantes dos resíduos de polpas de frutas tropicais: acerola (Malpighia glabra L.), goiaba (Psidium Guayaba L.), abacaxi (Ananas comosus L.), cupuaçu (Theobroma grandiflorum), bacuri (Platonia insignis) e graviola (Annona muricata L.). Os resultados encontrados mostraram que os resíduos analisados apresentaram quantidades significativas de macronutrientes (carboidratos, proteínas e lipídios). Todos os resíduos avaliados, com exceção do cupuaçu, apresentaram valores elevados de vitamina C. Quanto aos carotenóides, destacou-se o resíduo de acerola com 881,56 ± 9,01 µg/100 g e o resíduo de goiaba, com 644,9 ± 10,02 µg/100 g. Os resíduos analisados apresentaram baixas concentrações de antocianinas e flavonóides. Com relação aos teores de fenólicos totais se destacou o resíduo da polpa de acerola com 247,62 ± 2,08 mg/100 g. Portanto, pode-se concluir que os resíduos de polpas de frutas empregados neste estudo são fontes potenciais de macronutrientes e compostos bioativos, destacando-se os resíduos de acerola e goiaba como mais ricos em compostos antioxidantes.


Brazil is one of the largest agro-industrial residues producers, such as waste fruit pulp industries, which has contributed to the increased production of organic waste, causing serious environmental problems. In this context, studies have been conducted in order to investigate the nutritional value of these wastes, valuing them and suggesting new alternatives for use. The objective of this study was to characterize and determine nutritional antioxidant compounds of the waste pulp of the following tropical fruits: acerola (Malpighia glabra L.), guava (Psidium Guayaba L.), pineapple (Ananas comosus L.), cupuaçu (Theobroma grandiflorum), bacuri (Platonia insignis) and soursop (Annona muricata L.). The results showed that the waste analyzed showed significant amounts of macronutrients (carbohydrates, proteins and lipids). All waste assessed, except cupuaçu, showed elevated levels of vitamin C. As for carotenoids, highlighted the residue of acerola with 881.56 ± 9.01 µg/100 g and residue of guava, with 644.9 ± 10.02 µg/100 g. The waste also showed low concentrations of anthocyanins and flavonoids. Regarding of total phenolic levels, the residue of the pulp acerola with 247.62 ± 2.08 mg/100 g of total phenolics was high. Therefore, it may be concluded that the waste of fruit pulp used in this study are potential sources of nutrients and bioactive compounds, especially the waste from acerola and guava as richer in antioxidant compounds.
